Intestinal homeostasis and the coordinated actions of digestion, absorption and excretion are tightly regulated by a number of gastrointestinal hormones. Most of them exert their actions through G-protein-coupled receptors. Recently, we showed that the absence of Gαq/Gα11 signaling impaired the maturation of Paneth cells, induced their differentiation toward goblet cells, and affected the regeneration of the colonic mucosa in an experimental model of colitis. Although an immunohistochemical study showed that Gαq/Gα11 were highly expressed in enterocytes, it seemed that enterocytes were not affected in Int-Gq/G11 double knock-out intestine. Thus, we used an intestinal epithelial cell line to examine the role of signaling through Gαq/Gα11 in enterocytes and manipulated the expression level of Gαq and/or Gα11. The proliferation was inhibited in IEC-6 cells that overexpressed Gαq/Gα11 and enhanced in IEC-6 cells in which Gαq/Gα11 was downregulated. The expression of T-cell factor 1 was increased according to the overexpression of Gαq/Gα11. The expression of Notch1 intracellular cytoplasmic domain was decreased by the overexpression of Gαq/Gα11 and increased by the downregulation of Gαq/Gα11. The relative mRNA expression of Muc2, a goblet cell marker, was elevated in a Gαq/Gα11 knock-down experiment. Our findings suggest that Gαq/Gα11-mediated signaling inhibits proliferation and may support a physiological function, such as absorption or secretion, in terminally differentiated enterocytes.  相似文献

We have developed a new method for the large scale preparation of pyridylaminated (PA-) oligosaccharides from glycoproteins. Phenol/chloroform extration was adapted for the removal of protein and excess 2-aminopyridine, improving the efficiency of preparation. From a 2.5 g sample of human apo-transferrin, 25–30 mol of agalacto biantennary PA-oligosaccharide could be obtained. We have isolated a human cDNA clone encoding a novel acidic protein of MW 55,000 that we designated “myocilin” since it has homology to myosin and is localized preferentially in the ciliary rootlet and basal body of the connecting cilium of photoreceptor cells. The deduced amino acid sequence of human myocilin showed significant homologies with nonmuscle myosin ofDictyostelium discoideumin the N-terminal region and also with olfactomedin of bullfrog in the C-terminal region. Myocilin contained a leucine zipper-like motif similar to that seen in kinectin and other cytoskeletal proteins. These findings suggest that myocilin is a novel cytoskeletal protein involved in the morphogenesis of ciliated neuroepithelium such as photoreceptor cells. The myocilin gene (MYOC) was mapped to human chromosome 1q23–q24 by fluorescencein situhybridization.  相似文献

Ulex europaeus agglutinin-I (UEA-I) recognizes the Fuc alpha 1----2 Gal linkage. Receptors for UEA-I were not detected in mouse embryos until the 13th day of embryo-genesis, except for their temporary expression in early trophectoderm cells. In adult mice, UEA-I receptors were detected at various sites, including cells of the digestive tracts, the bronchial epithelium, Hassall's corpuscle of the thymus, and the skin. The fucose-binding protein of Lotus tetragonolobus (FBP) is another lectin that recognizes fucosyl residues. The distribution of FBP receptors was significantly different from that of UEA-I receptors. FBP receptors were first detected in late 8-cell embryos and were expressed in the embryonic ectoderm, visceral endoderm, and trophoblastic giant cells in egg-cylinders. At later stages, the distribution of FBP receptors became restricted to certain parts of the embryo. In the adult, the distribution of FBP receptors was more restricted than that of UEA-I receptors. Particularly in embryos before the 11th day of gestation, the distribution of FBP receptors resembled that of SSEA-1, which is defined by the Gal beta 1----4(Fuc alpha 1----3) GlcNAc linkage. From the specificity of FBP, we inferred that the disappearance of SSEA-1 and FBP receptors during embryogenesis is not the result of alpha 1----2 fucosylation of the terminal galactosyl residue in the determinant. The fact that the expression of two fucose-related cell-surface markers, i.e., UEA-I receptors and SSEA-1 (or FBP receptors), is developmentally regulated in an entirely different fashion is an excellent example illustrating the precise control of differentiation-dependent alterations in cell-surface carbohydrates.  相似文献

It was found that a non-tumorigenic epithelial cell line from the liver of a Buffalo-strain rat (BRL) secreted into the culture medium various inhibitors of the growth of BRL and RSV-BRL (tumorigenic BRL transformed by infection of Rous sarcoma virus). The secreted inhibitors were classified into two types: one inhibited the growth of BRL to a greater extent than that of RSV-BRL (non-tumorigenic BRL growth inhibitor, NGI), and the other, vice versa (tumorigenic BRL growth inhibitor, TGI). Two NGI (NGI-I and NGI-II) and two TGI (TGI-I and TGI-II) were highly purified from the serum-free conditioned medium. In sodium dodecyl sulfate-polyacrylamide gel electrophoresis without 2-mercaptoethanol, NGI-I and II gave protein bands with molecular weights (Mr) of 56,000 and 21,000, respectively. TGI-I and II gave a band that migrated faster than bromophenol blue marker dye, but they did not pass through an ultrafiltration membrane with an Mr cutoff of 5,000. In the presence of a reducing reagent, only NGI-II showed a decrease of Mr, from 21,000 to 11,000. NGI and TGI showed 50% growth inhibition with BRL and RSV-BRL, respectively, at 5-15 ng/ml in the medium containing 10% fetal calf serum. NGI and TGI all were stable to 1 M acetic acid (pH 2.3) and 6 M urea, but labile to 5 mM dithiothreitol or trypsin. Of the eight cell lines tested, NGI-I was most effective on BRL, NGI-II on BRL and HSC-3 (human tongue squamous carcinoma), and both TGI-I and II on RSV-BRL.  相似文献

A new species of the genus Stathmopoda from the Ryukyu Islands, Japan, is described. The new species is associated with Diospyros maritima Blume and is very similar to S. masinissa Meyrick, which is a notorious pest of persimmon, D. kaki L., in Japan and Korea. The adult external features, wing venation, and male and female genitalia of both species are illustrated and compared in detail.  相似文献
